So I want to do a DWN, BNE, TVL Return JASA.

It leaves DWN around lunch, gets to brassy in the arvo and up to TVL in the evening.

A quick overnight in TVL, departing at 6am and so on back home to DWN via Brissy.

All well and good when I want to go to TVL, but this next trip needs to put me in Brissy for the day, so the question is, how can I do this for the same points, but giving me a day in between in Brissy? Either on the way there or back.

Put simply, you cannot break it up, unless you want to take it as TSV BNE for 33k, and BNE DRW for 33 k

Any chance that the plat desk could defer the return BNE-DWN to the next day?
Unlikely.

If you choose the earliest TSV-BNE flight you can, you 'may' be able to get the BNE-DRW flight moved to a later time (not day, just time). This will depend if there is any U-bucket availability on the flight you want to move to. Then you would need to ask the Plat desk to change this leg. I don't know the flight times so this may not be feasible for you.

If I were to "miss" the Brisbane to Darwin return leg, would they put me on the following days flight? Or the next available if the same seat bucket was free?

Qantas & Jetstar Any Seat Award: If this is a Qantas & Jetstar Any Seat Award and the itinerary has been partially flown, then a proportion of the money and points originally used to obtain the award will be refunded to the applicable Member. The proportion refunded will be the remaining ticket value as allocated by Qantas as a proportion of the original ticket value.

Depends, on many different factors that may or may not be conveyed to you. What booking class is your JASA and what are its refund conditions? The big factor would be availability in the seat class and whether or not the person staffing the desk is allowed to rebook you.

It would be a big risk and you might find yourself stuck in a city you cannot get out of easily for a few days.

Hmmm stuck in BNE or DRW? Both different forms of hell.

Business Class, just looked at the fare conditions for the ASA.

I am based in Darwin, so there isnt a great issue with getting back here

As a regular on 824/825 I cannot see it happening, there just wont be the availability of the fare bucket next day, they often go three to four weeks out at the best of times.

In that case, would they put me in Y, with a comped J earn?

Alternatively, I could just go a no show on the BNE - TVL - BNE, in which case they would refund that leg.

The issue is, that the JASA to DWN-BNE-TVL-BNE-DWN is cheaper that the DWN-BNE-DWN

If you did no show to TSV, and then on return, the leg to Darwin would more than likely cancel. Not worth the risk, if you really want to go to Brisbane you need to pay the points or go via TSV I am afraid.
