Upgrade using points booked using amex credit

Is it possible to upgrade a fare booked on amex portal (using my $400 travel credit) and upgrade using points to J?? This is assuming we have space available. The route is BNE - CBR
 
Yes. I have done this.

Previously I was able to upgrade as soon as the E ticket came through as there was business available.

With my recent booking there was no business available so have put in a request for upgrade.
 
Yes you can, as above. You need to wait for the flight to be ticketed. I did this the other day, then jumped on QF booking page and initially it said "fare not eligible for upgrade" I nearly died. Waited for the flight to be ticketed the next day and checked again, put in the upgrade request.

What does that mean “flight to be ticketed”?

The booking needs to be ticketed by the airline (ie: a receipt is issued with ticket number) before an upgrade request should be submitted to QF. This is almost always within a few minutes - specially for domestic flights and via Amex.
What is J?
J is shortcut for Business Class. J being the letter associated with full fare Business Class bookings with QF.
