Which would be best? Buying tickets directly with American Airlines or using reward flight points with Qantas on same flight? Is there a difference in benefits e.g. points and status ,lounge etc.

It depends on your personal choice and price of the ticket. Qantas points are worth about 1.6c per point as per point hack.
FYI: If it is a short haul ticket, you might get better point redemption with avios.

It depends what you *could* use the points elsewhere for?

I use QFFF points for some fares which would otherwise be extremely expensive with cash. So I compare the price of a domestic USA with whatever else I need the points for.

If you have no other use for the points and want to save some cash, no issue using them.
 
Concurring with the previous ones. To give an example, I just checked the cash price and reward availability for an intra-European return flight. The points would have given 1.3c value to me which I deemed too low (using formula [(cash price - reward ticket fees) / points used] ). I'm opting to save those points for a higher value redemption and pay cash for these tickets.

It comes down to how much do you pay per point to earn them and where else could you redeem them.
