Hi, am flying on one ticket, Perth to Syd via QF and then out to Vancouver via Air Canada, and then back on Air Canada from Anchorage to Vancouver and then to Sydney and trans to QF for domestic flight back to Perth. They are interline airlines. I am getting confused as to whether we can use the QF seamless transfer for either direction. Time is tight so would be a bonus if we could. Does just one flight qualify you or does it have to be both flights?

Not sure what you mean by 'qualify you'.

Yes, on the same ticket your bags should be able to be tagged all the way from PER to YVR and from ANC to PER (I do QF-AC and AC-QF connections fairly regularly).

Don't forget Canada is soon moving to a ESTA type of e-approval before you fly - like ESTA you have to apply on line. And Canadian security immigration can be time consuming like the USA. Are you transiting Canada in your ANC-YVR-SYD?

Yes, one flight to or from SYD entitles you to use the service.
You will have to have had your bags, if any, checked through from Perth on the way out.

By "qualify" I merely meant were we entitled to use the transfer or did we have to look elsewhere for separate inter-terminal transfer. Looks like we have to collect our bags in Sydney before domestic leg to Perth. Yep, we have our Canadian documentation. We are not stopping over in Canada on way home, just a connecting flight.

You always have to collect your bags on arrival into Australia regardless of any onward domestic bookings.
However you should be able to check in at the transfer desk in the international terminal, drop your bags and then catch the airside bus to the domestic terminal.
HOWEVER don't get too excited. It isn't as quick and easy as it sounds.

Just to be clear:

Outbound from PER:

Check bags all the way - you won't see them until arrival in YVR.
When you ger to SYD T3, head to gate 15 where your 'seamless' bus to International T1 awaits. Itwill drop you off landside. (Make sure you keep your PER-SYD BP's as these prove your entitlement you to use the service.)

Inbound to PER:

On arrival into T1, you get process by immigration, collect bags and get process by customs. You then head to the far eastern end of T1 where the Qantas 'Seamless' facility building is (this is landside). There you recheck your bags, before going through security to wait for a bus to take you to T3 (gate 15) and your flight to PER.
