We have flights PER SIN. 12 hours later SIN TYO, 10 hours transit hotel booked. Can we check in suitcase from PER SIN TYO

Yes Just tell them at check in you want it booked all the way

Yes, I believe they will through check it, if onward transit is within 24 hours. I had a 18 layover once and it was held and checked through. Not sure where they stored it though as the bag was water damaged on subsequent collection.

Can this still be done if the two flights are on different bookings? Say MEL to SIN on one booking and SIN to MLE on another.

Yes, they will do it. My transit was 23h 30m and they could do it.

What I did not realise was that you cannot check in 2 bags to 2 different destinations - i.e. 1 bag to SIN and another to your final destination. All bags need to go together.

Yes, I did PER-SIN-SEA back in March with an overnight at YOTELAIR Changi. Bag at Perth was checked all the way through to US leaving the next morning. Just put things I needed overnight into my carry-on.

Coming back flew SFO-SIN-PER with two nights again at Changi. Checking in at SFO the person was originally going to check my bag all the way through to PER. Since I had two nights in SIN I requested they only check it to SIN. Interestingly, the check-in staff had to get a supervisor to assist as she didn't know how to only check the bag to first stop.

All baggage check-in went smoothly in both directions. As you're packing, just consider what you will need in-transit and remember the 100ml liquids rules. In a previous trip through SIN I accidentally had a 150ml lotion container in carry-on but it was only about half full. The co-operative security staff actually provided an empty 100ml plastic bottle for me to use so that I was within limits and could still carry the lotion.
