What I can see on our Routemap is that Qantas only offer flight to Dubai from Manchester, do not offer anyother cities out of Manchester

My dates are flexible but would like to start my holiday around 23rd May 2017 and return to Aus around 20th June, with a stopover in different destinations both ways, for around 2 to 3 days.
 
If you want to stopover in South America (e.g. Santiago) on the return, you will probably need to book through a travel agent. This can't be booked online.

You can stopover in Dubai on the outbound. You could fly Brisbane-Sydney-Dubai(stopover)-London-Manchester. By flying from Sydney to London, you would also be able to choose Qantas-operated flights all the way to London. This is important if you want to upgrade with points. You can fly Brisbane-Dubai-Manchester on Emirates, but you won't be able to upgrade using Qantas points on either flight.

For the return, you could fly home via the USA (e.g. New York, Dallas, San Francisco or Los Angeles), or via somewhere in Asia (e.g. Hong Kong, Bangkok or Singapore). This can easily be booked using the multi-city booking tool on the Qantas website.

If returning via the USA, you'll likely need to fly on BA or AA from London to a city where Qantas flies, then pick up the Qantas flight back to Australia. If returning home via Asia, you could take Qantas to Dubai, then Emirates to an Asian destination that Qantas flies to. Then you can use Qantas for the last leg back to Australia.

FWIW, returning home via Asia will be cheaper than the Americas.

I was able to price this on the Qantas website:

I would note that these are sale fares though, so not upgradeable. If you wanted to be able to upgrade, you would need to purchase Saver fares. For this itinerary, that would cost ~$2,900. You'd need to either book through a travel agent, or wait for the current Qantas sale to finish and book online.
 
This itinerary looks ideal for what we have in mind, will have a closer look at it thanks Matt.
 
The 12 hour flight from London to LA with BA might be a bit of a long haul with no prospect of an upgrade though:)

Just to be clear, none of the flights on this ticket would be upgradeable as they are Sale fares. You would, at a minimum, need to purchase "Saver" tickets. Then at least the Qantas flights are upgradeable.

FWIW, you should also be able to use New York as a US stopover. The BA flight from London to New York is much shorter than London-LA. They you could fly Qantas from New York to Brisbane via Los Angeles. ;)
