Can a FHR reservation made using the Supplementary Card entitled to complimentary benefits include guaranteed 4pm late check-out, room upgrades (subject to availability), daily breakfast for two, or a complimentary amenity valued at US$100 as well.

Yes, when the supp cardholder is logged into their own Amex account they will be able to book FHR rates via Amex Travel.

Good to know. I do have another personal card as an AC holder in my name under the same account. Can I use that to booked. My whole intention is to book one night using the the primary card and another night with the AC card or the supplementary card. Wonder if I could have the USD200 Hotel credit.
 
Back-to-back stays booked by a single Card Member, Card Members staying in the same room or Card Members traveling in the same party within a 24-hour period at the same property are considered one stay and are ineligible for additional FHR benefits ("Prohibited Action"). American Express and the Property reserve the right to modify or revoke FHR benefits at any time without notice if we or they determine, in our or their sole discretion, that you may have engaged in a Prohibited Action, or have engaged in abuse, misuse, or gaming in connection with your FHR benefits.

To answer the question, yes you can select between your primary and supp AC card, both appear with FHR access when logging into Amex Travel. But as discussed it might not be a great idea to do back to back stays unless you have a card in your travelling partner's name and switch between yours and theirs.

I knew back to back by the same person would count as one stay. I've had success booking one night in my name and the second night in MrsDaver6's name and getting a $200 credit. IIRC though I think we needed to use $100 for each "stay". No room change required.
